A direct LED replacement yes, but you’ll probably need to play around with centering the LED in the reflector unless you get a compatible spacer, like from here: Kaidomain Misc Parts
I’ve had success with utilizing a larger spacer (eventually by chance the LED gets centered), but it certainly is easier and looks better with the correct spacer.

There I go again —Not comprehending what I read in the OP —— SST40 in my opinion in a S2 makes a terrible beam —but in all fairness I really don’t care for a XM-L2 in them either
If the SST-40 MCPCB is also 16mm, yes.

You might want to upgrade the driver as well to get the most out of it.
BG sells the BLF X6 driver, which is an interesting replacement.
Edit, boy these drivers are expensive at BG nowadays, they used to be 8USD or so.

This is a 2-year-old unit that I had in my spares box.
LED is already dead due to some mishandling of the module.
Having been exposed to other lights these couple of of years, I now intend to make this a working light again.
So I’m taking it easy — LED mcpb at the very least.

If you don’t have a Nichia 219B flashlight yet, I would suggest sticking one of these beauties in there: 219B R9080 LED
